Duchenne's muscular dystrophy (DMD), which affects 1/3500 live male births, involves a progressive degeneration of skeletal and cardiac muscle, leading to early death. The protein dystrophin is lacking in DMD and present, but defective, in the allelic, less severe, Becker muscular dystrophy and is also missing in the mdx mouse. Experiments on the mdx mouse have suggested two possible therapies for these myopathies. Implantation of normal muscle precursor cells (mpc) into mdx skeletal muscle leads to the conversion of dystrophin‐negative fibres (...) to ‐positive, with consequent improvement in muscle histology. Direct injectidn of dystrophin cDNA into skeletal or cardiac muscle also gives rise to dystrophin‐positive fibres. Although both appear promising, there are a number of questions to be answered and refinements to be made before either technique could be considered possible as treatments for myopathies in man. Catholic theology’s traditional understanding of the spiritual nature of the human person begins with the idea of a rational soul and human mind that is made manifest in free will—the spiritual experience of the act of consciousness and cause of all human arts. The rationale for this religion-based idea of personhood is key to understanding ethical dilemmas posed by modern research that applies a more empirical methodology in its interpretations about the cause of human consciousness. Applications of these beliefs about (...) the body/soul composite to the theory of evolution and to discoveries in neuroscience, paleoanthropology, as well as to recent animal intelligence studies, can be interpreted from this religious and philosophical perspective, which argues for the human soul as the unifying cause of the person’s unique abilities. Free will and consciousness are at the nexus of the mutual influence of body and soul upon one another in the traditional Catholic view, that argues for a spiritual dimension to personality that is on a par with the physical metabolic processes at play. Therapies that affect consciousness are ethically problematic, because of their implications for free will and human dignity. Studies of resilience, as an example, argue for the greater, albeit limited, role of the soul’s conscious choices in healing as opposed to metabolic or physical changes to the brain alone. (shrink)

Ancient DNA has been discovered in many types of preserved biological material, including bones, mummies, museum skins, insects in amber and plant fossils, and has become an important research tool in disciplines as diverse as archaeology, conservation biology and forensic science. In archaeology, ancient DNA can contribute both to the interpretation of individual sites and to the development of hypotheses about past populations. Site interpretation is aided by DNA‐based sex typing of fragmentary human bones, and by the use of genetic (...) techniques to assess the degree of kinship between the remains of different individuals. On a broader scale, population migrations can be traced by studying genetic markers in ancient DNA, as in recent studies of the colonisation of the Pacific islands, while ancient DNA in preserved plant remains can provide information on the development of agriculture. Significant life events tend to cause transformational change, but most narratives in agriculture focus on how negative life events such as death, bankruptcy, and health problems have created change. International experiences can be positive, significant life events. Transformational tourism has been shown to change travelers’ behavior. Therefore, we propose to examine the perceived outcomes of international experiences by agricultural leadership alums. Unstructured interviews with 36 agricultural leadership alums from IFYE, Nuffield, and LEAD Nebraska with distinctive international experiences were interviewed. The (...) international experiences varied in purpose, time abroad, cultural immersion, structured travel, and independent study. This study answers the following questions: (1) How does international travel for producers lead to transformation? (2) How does international travel support behavior change? Data show the international experiences were impactful and transformative due to two processes that occurred during the international experience. First, agricultural leadership participants experienced disorienting events that forced them to reconsider and reject their current mental models of the agricultural system. Second, experiential learning through visiting farms, meeting producers, and spending time reflecting on what they were experiencing with their peers allowed the participants to rebuild their mental models and be inclusive of the broader agricultural system. During these two processes, the participants’ attitudes, subjective norms, and perceived behavior control of new agricultural practices, sustainable agriculture, and possible options within the agricultural system expanded. Short-term international experiences were an important gateway to further international experiences. Longer-term international experiences that involved independent study, cultural immersion, and global network development supported the adoption of sustainable agriculture through perceived attitudes, social norms, and behavioral control. To support transformational change in the agricultural system, we need as many tools as possible to support practitioners. International experiences provide a tool to support transformational change. It has been shown that visual awareness in the blind hemifield of hemianopic cats that have undergone unilateral ablations of visual cortex can be restored by sectioning the commissure of the superior colliculus or by destroying a portion of the substantia nigra contralateral to the cortical lesion (the Sprague effect). We propose that the visual awareness that is recovered is due to synchronized oscillatory activities in the superior colliculus ipsilateral to the cortical lesion. These oscillatory activities are normally partially suppressed (...) by the inhibitory, GABAergic contralateral nigrotectal projection, and the destruction of the substantia nigra, or the sectioning of the collicular commissure, disinhibits the collicular neurons, causing an increase in the extent of oscillatory activity and/or synchronization between activities at different sites. This increase in the oscillatory and synchronized character is sufficient for the activities to give rise to visual awareness. We argue that in rodents and lower vertebrates, normal visual awareness is partly due to synchronized oscillatory activities in the optic tectum and partly due to similar activities in visual cortex. It is only in carnivores and primates that visual awareness is wholly due to cortical activities. Based on von Baerian recapitulation theory, we propose that, even in humans, there is a period in early infancy when visual awareness is partially due to activities in the superior colliculus, but that this awareness gradually disappears as the nigrotectal projection matures. Based on theoretical considerations of Aurell (1979) and Block (1995), we argue that object recognition awareness is distinct from purely sensory awareness and that the former is mediated by neuronal activities in areas that are separate and distinct from cortical sensory areas. We propose that two of the principal functions of neuronal activities in sensory cortex, which are to provide sensory awareness and to effect the computations that are necessary for object recognition, are dissociated. We provide examples of how this (...) dissociation might be achieved and argue that the components of the neuronal activities which carry the computations do not directly enter the awareness of the subject. The results of these computations are sparse representations (i.e., vector or distributed codes) which are activated by the presentation of particular sensory objects and are essentially engrams for the recognition of objects. These final representations occur in the highest order areas of sensory cortex; in the visual analyzer, the areas include the anterior part of the inferior temporal cortex and the perirhinal cortex. We propose, based on lesion and connectional data, that the two areas in which activities provide recognition awareness are the temporopolar cortex and the medial orbitofrontal cortex. Activities in the temporopolar cortex provide the recognition awareness of objects learned in the remote past (consolidated object recognition), and those in the medial orbitofrontal cortex provide the recognition awareness of objects learned in the recent past. The activation of the sparse representation for a particular sensory object in turn activates neurons in one or both of these regions of cortex, and it is the activities of these neurons that provide the awareness of recognition of the object in question. This book explores the principal issues involved in bridging the gap between the pure theory of consumer behavior and its empirical implementation. The authors focus upon the structure of preferences, the treatment of demographic variables, the treatment of dynamics, and the specification of the stochastic structure of the demand system.

Recent experiments have shown that the amplitudes of cortical gamma band oscillatory activities that occur during anesthesia are often greater than amplitudes of similar activities that occur without anesthesia. This result is apparently at odds with the hypothesis that synchronized oscillatory activities constitute the neural correlate of consciousness. We argue that while synchronization and oscillatory patterning are necessary conditions for consciousness, they are not sufficient. Based on the results of a binocular rivalry study of Fries et al. (1997), we propose (...) that the degrees of oscillatory strength and synchronization of neuronal activities determine the degree of awareness those activities produce. On the other hand, the overal firing rates of neurons in cortical sensory areas are not correlated with the degree of awareness the activities of those neurons produce. The results of the experiment of Fries et al. (1997) appear to conflict with the results of another binocular rivalry experiment, in which monkeys were trained to pull a lever in order to report which stimulus object was being perceived (Leopold & Logothetis, 1996). In the latter experiment, it was demonstrated that the firing rates of neurons in striate cortex did not change during perceptual alterations, while 90% of neurons in inferior and superior temporal cortices changed their firing rate when the perceived image changed. This result led to the conclusion that activities in temporal cortex are correlated with visual awareness, but those in striate cortex are not. We argue that activities in temporal cortex contribute little, if anything, to perceptual awareness, and that their primary function is computational. Thus the correlation between the firing rates of neurons in these areas and the responses of the monkeys is due to the recognition of a particular stimulus object, which in turn is due to the computations made there. The neural and endocrine bases of the generation of thirst are reviewed. Based on this review, a hierarchical system of neural structures that regulate water conservation and acquisition is proposed. The system includes primary sensory-receptive areas; secondary sensory structures (circumventricular organs), which detect levels of hormones, including angiotensin II and vasopressin, which are involved in generating thirst; preoptic and hypothalamic structures; and an area within the ventrolateral quadrant of the periaqueductal gray matter. Hodological and other data are used to determine (...) the hierarchical organization of the system. Based on studies of the effects of lesions to various structures within the hierarchy of the system, it is proposed that the awareness of thirst in rodents is either entirely or predominantly due to neuronal activities in a subsection of the ventrolateral periaqueductal gray matter. It is also hypothesized that the awareness of thirst in primates is due to neuronal activities in both the ventrolateral periaqueductal gray and in a region within the medial prefrontal and anterior cingulate cortex. Genome‐wide identification of transcription factor binding sites with the ChIP‐seq method is an extremely important scientific endeavor − one that should ideally be performed for every transcription factor in as many cell types as possible. A major hurdle on the way to this goal is the necessity for a specific, ChIP‐grade antibody for each transcription factor of interest, which is often not available. Here, we describe CETCh‐seq, a recently published method utilizing genome engineering with the CRISPR/Cas9 system to circumvent the (...) need for a specific antibody. Using the CETCh‐seq method, targeted genomic editing results in an epitope‐tagged transcription factor, which is recognized by a well‐characterized, standard antibody, efficacious for ChIP‐seq. We have used CETCh‐seq in human cancer cell lines as well as mouse embryonic stem cells. We find that roughly 60% of transcription factors tagged using CETCh‐seq produce a high quality ChIP‐seq map, a significant improvement over traditional antibody‐based methods. (shrink)

The 6th Consensus Statement on Concussion in Sport is authored by the Concussion in Sport Group (CiSG) and intends to provide evidence-based recommendations on concussion management for the welfare of sports participants. However, the authors of the Consensus Statement have declared many competing links to third-party groups. While the declaration of an author’s competing interests is now a widely accepted practice within academic publishing aimed at greater transparency and research integrity, it is not a measure to remove the potential influence (...) of third-party links. In the case of the Consensus Statement, this leaves uncertainty about how the potential sources of third-party bias declared by individual members of the CiSG may have influenced the CiSG’s interpretation of the existing state of evidence on concussion management and impacted the content of the recommendations contained with the Consensus Statement. Openly declaring third-party links falls short of securing trustworthiness in the Consensus Statement recommendations since it leaves end-users of the Consensus Statement with the task of interpreting the impact(s) of these competing interests with little information to guide them. This is because the authors themselves offer neither explanation nor guidance concerning potential bias arising from the competing interests that they have declared, which impinges upon the ability of readers to evaluate the trustworthiness of the content. The status quo could be improved by excluding researchers who have conflicts of interest. Nevertheless, if authors with competing interests are permitted to be part of the CiSG (as is the status quo), then I propose a novel option for increasing transparency that would require authors of the Consensus Statements to provide a reflexive explanation, not simply a declaration, of their competing interests. The religiously committed, for example, regularly assemble to engage in religious rites, including corporate liturgical worship. Although the participation in liturgy is central to the religious lives of many, few philosophers have given it attention. In this book Terence Parsons revives the older tradition of taking such objects at face value. Using various modern techniques from logic and the philosophy of language, he formulates a metaphysical theory of nonexistent objects. The theory is given a formalization in symbolism rich enough to contain definite descriptions, modal operators, and epistemic contexts, and the book includes a discussion which relates the formalized theory explicitly to English.

This extended investigation of the semantics of event (and state) sentences in their various forms is a major contribution to the semantics of natural language, simultaneously encompassing important issues in linguistics, philosophy, and logic. It develops the view that the logical forms of simple English sentences typically contain quantification over events or states and shows how this view can account for a wide variety of semantic phenomena. Focusing on the structure of meaning in English sentences at a &"subatomic&" level&-that is, (...) a level below the one most theories accept as basic or &"atomic&"&-Parsons asserts that the semantics of simple English sentences require logical forms somewhat more complex than is normally assumed in natural language semantics. His articulation of underlying event theory explains a wide variety of apparently diverse semantic characteristics of natural language, and his development of the theory shows the importance of seeing the distinction between events and states. Parsons demonstrates that verbs, also, indicate kinds of actions rather than specific, individual actions. Verb phrases, too, he argues, depend on modifiers to make their function and meaning in a sentence specific. An appendix gives many of the details needed to formalize the theory discussed in the body of the text and provides a series of templates that permit the generation of atomic formulas of English. Adolescents present a puzzle. There are foundational unclarities about how they should be regarded as decision-makers. Although superficially adolescents may appear to have mature decisional capacity, their decision-making is in many ways unlike that of adults. Despite this seemingly obvious fact, a concern for the claims of autonomy has led to the development of the legal doctrine of the mature minor. This legal construct considers adolescents, as far as possible, as equivalent to adults for the purpose of medical decision-making. The (...) movement to support independent decision-making by adolescents through providing information to them and securing their consent apart from their parents is encouraged by those legal understandings that hold that unemancipated minors should generally be considered as possessing effective decisional capacity. Such legal structures, however, do not adequately take account of the wide variations in adolescent capacities, the immaturity of most adolescent decision-makers, or the important contributions made by parents to the development of their adolescents through parental partnering in the adolescent’s decision-making. The data available indicate that in general adolescents should be regarded as apprentice decision-makers who should make decisions in collaboration with their parents until at least the age of 18. Steps should not be taken pre-emptively to isolate adolescents from the guidance of their parents. As a general rule, what Piker has referred to as “collaborative paternalism” appears most likely both to protect adolescents from their own untoward choices, while also very importantly helping them with parental guidance to develop into mature decision-makers with the capacity to make medical choices on their own. Since the early 1980’s, with the clinical advent of in vitro fertilization resulting in so-called “test tube babies,” a wide array of ethical considerations and concerns regarding artificial womb technology (AWT) have been described. Recent breakthroughs in the development of extracorporeal neonatal life support by means of AWT have reinitiated ethical interest about this topic with a sense of urgency. Most of the recent ethical literature on the topic, however, pertains not to the more imminent scenario of a physiologically improved (...) method of neonatal care through AWT, but instead to the remote scenario of “complete ectogenesis” that imagines human gestation occurring entirely outside of the womb. This scoping review of the ethical literature on AWT spans from more abstract concerns about complete ectogenesis to more immediate concerns about the soon-to-be-expected clinical life support of what we term the fetal neonate or fetonate. Within an organizing framework of different stages of human gestational development, from conception to the viable premature infant, we discuss both already identified and newly emerging ethical considerations and concerns regarding AWT and the care of the fetonate. In this paper I attempt a moral justification of protecting wild species, ecosystems, and landscapes, a justification not directly grounded in appeals to human benefit. I begin with a description of anthropocentric and ecosystemic approaches to the valuing of nature and offer some empirical arguments in support of the ecosystemic view. I suggest that human beings have a genetic need for natural environments, and that the direct experience of wild nature is an intrinsic good. Theoretical coherence and scope is another (...) advantage of the ecological perspective over the anthropocentric view. Turning to moral psychology, I argue that human beings have a fundamental need to care for things outside themselves and that this need is suitably met, and human life enriched, by a transcending concern for the well-being of natural species, habitats, and ecosystems . These considerations are joined with the ecological point of view to yield the conclusion that a self-transcending concern for the welfare of wild species and their habitats enriches the quality of moral life. Persons with genuine reverence and respect for wild creatures and their habitats will enjoy greater fulfilment in their own lives and be better neighbors toeach other. (shrink)

The authors of Austere Realism describe and defend a provocative ontological-cum-semantic position, asserting that the right ontology is minimal or austere, in that it excludes numerous common-sense posits, and that statements employing such posits are nonetheless true, when truth is understood to be semantic correctness under contextually operative semantic standards. Terence Horgan and Matjaz [hacek over z] Potrc [hacek over c] argue that austere realism emerges naturally from consideration of the deep problems within the naive common-sense approach to truth (...) and ontology. They offer an account of truth that confronts these deep internal problems and is independently plausible: contextual semantics, which asserts that truth is semantically correct affirmability. Under contextual semantics, much ordinary and scientific thought and discourse is true because its truth is indirect correspondence to the world. After offering further arguments for austere realism and addressing objections to it, Horgan and Potrc [hacek over c] consider various alternative austere ontologies. They advance a specific version they call "blobjectivism"--the view that the right ontology includes only one concrete particular, the entire cosmos, which, although it has enormous local spatiotemporal variability, does not have any proper parts. The arguments in Austere Realism are powerfully made and concisely and lucidly set out. The authors' contentions and their methodological approach--products of a decade-long collaboration--will generate lively debate among scholars in metaphysics, ontology, and philosophy. Terence E. This paper explores the view that there are such things as (nonexistent) fictional objects, and that we refer to such objects when we say things like "Sherlock Holmes is a fictional detective", or "Conan Doyle wrote about Sherlock Holmes". A theory of such objects is developed as a special application of a Meinongian Ontology.

Thinking with Literature offers a succinct introduction to a cognitive literary criticsm, broad in scope but focusing on a particular cluster of approaches, some of which have so far been little used. Explanatory chapters and sections alternate with close readings of literary texts from a wide range of different periods and genres. The literary readings are not mere 'examples' of cognitive topics, still less of hypotheses in cognitive science: the central argument is that cognitive criticism must draw its primary energies (...) from the complexities of literature itself. A cross-cultural empirical study is reported in this article which looks at ethical beliefs and behaviours among French and German managers, and compares this with previous studies of U.S. and Israeli managers using a similar questionnaire. Comparisons are made between what managers say they believe, and what they do, between managers and their peers' attitudes and behaviours, and between perceived top management attitudes and the existence of company policy. In the latter, significant differences are found by national ownership of the (...) company rather than the country in which it is situated. Significant differences are found, for both individual managers by nationality, and for companies by nationality of parents, in the area of organizational loyalty. The attitude towards accepting gifts and favours in exchange for preferential treatment, as a measure of societal values, is also found to show significant differences between national groups. However, no significant differences are found for measures for group loyalty, conflict between organizational and group loyalty and for conflicts between self and group/organization. The findings have implications for cross-border management decision strategies regarding such issues as receiving and giving of gifts, and the management of relations between local employees and international organizations which may be affected by differences in attitude to corporate loyalty. The radical contingencies attending human reproduction indicate that attempts to improve the living conditions of future generations result in generations populated by different individuals than would otherwise have been born. This remarkable consequence challenges the widespread belief that the present generation has responsibilities to its remote successors. I contend, first, that while the radical genetic contingency and epistemological indeterminacy of future persons obsolves us of obligations to act "in behalf of" them as individuals, this moral absolution does not entail a (...) permission to disregard entirely the remote consequences of our policies. Since relevant moral principles bind us to persons in general, not to particular individuals, we remain obligated to improve the life prospects of whatever individuals eventually come into being. Second, I suggest that by applying an analogous argument within the lives of persons rather than to the long history of civilization, we arrive at the morally repugnant result of negating long-term obligations to contemporary persons. Conversely, the condition of continuity which affords moral legitimacy of personal obligations among contemporaries likewise entails moral responsibility for the life conditions of distant generations.
